Effect of omega -3 fatty acid diet on bone in healthy mice

Intake of polyunsaturated fatty acids (PUFA) can affect bone health by regulating calcium metabolism, prostaglandin synthesis, lipid oxidation, osteoblast formation, and osteoclastogenesis. The aim of the present study was to evaluate the effect of dietary polyunsaturated fatty acids on bone health in mice.
experimental method
Three-week-old male (n = 30) and female (n = 30)C57BL/6J mice were randomly divided into three feed groups. These feeds differed only in fat composition: soybean oil (soy), rich in omega -6 polyunsaturated fatty acids; docosahexaenoic acid (DHA) alone, an omega -3 polyunsaturated fatty acid; and DHA to arachidonic acid, an omega -6 polyunsaturated fatty acid, in a 20:1 ratio (DHA/ARA). After 9 weeks of feed feeding, the femur was taken for micro-computed tomography analysis and three-point bending mechanics test. Each group of mice was individually and serially bled to measure biomarkers of bone formation and bone resorption.
Experimental results
At the microstructural level, some parameters in cortical bone reached statistically significant differences in female mice, but these differences were too small to be considered biologically relevant. Similarly, there were statistical differences in trabecular bone parameters in male mice in some of the feed groups, but such subtle changes were difficult to interpret as diet-related.
There are subtle differences in the microstructure of bone, and these are likely the result of random effects that do not translate into biologically relevant changes. In conclusion, dietary intake of polyunsaturated fatty acids does not appear to affect bone structure or metabolism in healthy growing mice.
